With the visitors already trailing 2-0 in the series, fast bowler Nathan Ellis and young player Joel Davies have been ruled out of the concluding match, while five-wicket star Jack Edwards is under an injury cloud.
The Australians suffered a 32-run defeat in the second ODI, where their physical troubles compounded significantly as injuries piled up throughout the fixture.
Ellis managed just four overs before being forced off for the remainder of the South African innings, finishing with figures of 0/22 after taking three wickets in the series opener.
Meanwhile, Davies sustained a hamstring injury while substitute fielding during the second match, ending his hopes of featuring in the dead rubber.
Edwards delivered a standout performance by claiming five wickets to help contain South Africa's massive total of 365, but Cricket Australia confirmed he is also battling a hamstring issue.
The all-rounder travelled with the squad on the 90-minute bus journey from Johannesburg to Potchefstroom, though his availability for the final fixture remains uncertain.
